چکیده

A recent study showed an association between the dopamine D2 receptor gene (DRD2) and smoking. The purpose of this study was to determine if the familial transmission of smoking is linked to variation at the DRD2 locus in a genetically informative sample. Subjects were identified in alcohol treatment centers and their relatives were recruited for study. All subjects were interviewed to assess alcohol dependence, smoking habits, and psychiatric disorders. Two polymorphisms within the DRD2 gene were analyzed, including the TaqIA polymorphism. The sample consisted of 138 nuclear families with at least one offspring with habitual smoking, and analysis was by the transmission disequilibrium test (TDT), which avoids problems due to population stratification. There was no significant difference in the frequency between DRD2 alleles transmitted and not transmitted to habitual smokers. There also was no evidence for unequal transmission of DRD2 alleles for the phenotypes "ever smoker" or comorbid alcohol dependence and habitual smoking. This study does not support linkage of the DRD2 with smoking.

Association between the dopamine D2 receptor A2/A2 genotype and smoking behavior in the Japanese.

For the study presented here, we investigated possible links between the dopamine D2 receptor (DRD2) TaqIA genotype (DRD2*A) and smoking behavior in a total of 332 Japanese individuals. For the first time, functional insertion/deletion polymorphism (-141C Ins/Del) in the DRD2 promoter was also examined in relation to smoking behavior. The distribution of the DRD2*A genotype was significantly di...

A review of the role of dopamine receptors and novel therapeutic strategies in non-small cell lung cancer (NSCLC)

Lung cancer is a very aggressive and most deadly cancer in both men and women. Lung cancer is divided into two types of small cell lung cancer (SCLC) and non-small cell lung cancer (NSCLC). NSCLC is divided into 3 subgroups: adenocarcinoma (AC), squamous cell carcinoma (SqCC) and large cell carcinoma (LCC). Dopamine is involved in controlling motions, cognition, emotions, memory and reward mech...

Effects of Interaction Between Dopamine D2 Receptor and Monoamine Oxidase A Genes on Smoking Status in Young Men.

Although the effect of gene-gene interaction on nicotine-dopamine metabolism for smoking behavior has been reported, polymorphisms of dopamine D2 receptor (DRD2) and monoamine oxidase A (MAOA) have not been simultaneously examined among smokers. In this study, 481 young Taiwanese men completed a self-report questionnaire on smoking status, and data were obtained on polymorphisms of DRD2 rs18004...

Hyperprolactinemia and CYP2D6, DRD2 and HTR2C genes polymorphism in patients with schizophrenia

Introduction: Hyperprolactinemia is a common serious side effect of antipsychotic medications that are currently used in the treatment of patients with schizophrenia. Pharmacogenetic approaches offer the possibility of identifying patient-specific biomarkers for predicting the risk of this side effect. We investigated a possible relationship between variants (SNPs) in genes for cytochrome 2D6 (...

Association and family studies of DRD2 gene polymorphisms in alcohol dependence syndrome.

INTRODUCTION The human dopamine receptor 2 gene DRD2 plays a central role in susceptibility to Alcohol Dependence Syndrome (ADS). The aim of this study was to evaluate 3 single nucleotide polymorphisms: D2 (rs1076560), Tag1D (rs1800498), Tag1B (rs1079597) located in dopamine receptor 2 DRD2 gene and its role in alcohol dependence. MATERIAL AND METHODS DNA was provided from alcohol dependent (...
